Description

This therapeutic agent is a state-of-the-art Spiegelmer, consisting of a synthetically produced L-ribonucleic acid (L-RNA) aptamer conjugated to a polyethylene glycol (PEG) polymer. The use of a mirror-image L-RNA configuration renders the molecule exceptionally resistant to degradation by naturally occurring D-RNA specific nucleases, which dramatically enhances its in vivo stability and pharmacokinetic profile. The aptamer is engineered for high-affinity binding to Chemokine (C-C motif) ligand 2 (CCL2), effectively neutralizing its biological activity. By preventing CCL2 from engaging with its cognate receptor CCR2, this therapeutic blocks the key signaling pathway responsible for recruiting monocytes and macrophages to sites of inflammation and tissue damage. This targeted mechanism makes it a promising candidate for treating a range of chronic inflammatory and fibrotic diseases.

Chemokine (C-C motif) ligand 2 (CCL2), also widely known as Monocyte Chemoattractant Protein-1 (MCP-1), is a potent pro-inflammatory chemokine. It plays a pivotal role in the innate immune response by orchestrating the migration and infiltration of monocytes, memory T-lymphocytes, and natural killer (NK) cells. Secreted by a variety of cell types upon inflammatory stimulus, CCL2 exerts its effect by binding to the G protein-coupled receptor CCR2. The CCL2/CCR2 signaling axis is a fundamental pathway in the pathogenesis of numerous diseases characterized by chronic inflammation, including diabetic nephropathy, rheumatoid arthritis, atherosclerosis, and fibrosis.
