Target Overview

TAR DNA-binding protein 43 (TDP-43) is a highly conserved protein that primarily resides in the nucleus and is critically involved in RNA processing, including transcription, alternative splicing, and mRNA stability. TDP-43 is most notable for its central role in neurodegenerative diseases. In nearly all cases of Amyotrophic Lateral Sclerosis (ALS) and over half of the cases of Frontotemporal Lobar Degeneration (FTLD), TDP-43 becomes pathologically altered. It mislocalizes from the nucleus to the cytoplasm, becomes hyperphosphorylated and ubiquitinated, and aggregates into insoluble inclusions within neurons. This "proteinopathy" is a defining pathological hallmark of these diseases.
