This is an engineered, pegylated RNA aptamer that specifically binds to and neutralizes the 165-amino acid isoform of Vascular Endothelial Growth Factor (VEGF)-165. By preventing VEGF from binding to its cell surface receptors, this therapeutic agent inhibits abnormal blood vessel growth (neovascularization) and vascular permeability. This mechanism makes it a targeted treatment for neovascular (wet) age-related macular degeneration (AMD).

Target Overview

The vascular endothelial growth factor (VEGF) family comprises VEGF-A, VEGF-B, VEGF-C, VEGF-D, VEGF-E, and placental growth factor (PIGF). VEGF signaling through its receptors is fundamentally essential for angiogenesis. Since most tumors critically depend on angiogenesis, VEGF, the most potent cytokine driving this process, is central to tumor growth. Five distinct isoforms of VEGF-A arise from alternative splicing of a single gene. These isoforms bind to two specific tyrosine kinase receptors: VEGFR-1 (flt-1) and VEGFR-2 (flk-1/KDR). These receptors exhibit expression primarily restricted to endothelial cells.
