Nuclear factor erythroid 2-related factor 2 (Nrf2) is a transcription factor that serves as the master regulator of the cellular defense system against oxidative stress. Under normal conditions, Nrf2 is kept inactive in the cytoplasm by its inhibitor, KEAP1. In response to oxidative stress, Nrf2 is stabilized, moves into the nucleus, and activates the transcription of hundreds of antioxidant, detoxification, and cytoprotective genes. While this pathway is crucial for protecting against aging and chronic disease, it is often hijacked by cancer cells to promote their survival and resistance to chemotherapy.
