Description

The 9L/lacZ cell line was developed from the 9L cell line (a rat nitrourea-induced glioma cell line) in 1989. The 9L cell line was infected with a retroviral vector carrying E. coli encoding the beta-galactosidase lacZ gene and the G418-resistant Tn5 neomycin gene BAG replication defect. The cells were cultured for 14 days in the presence of G418, cloned, and tested for beta-galactosidase production. 9L/lacZ produces high levels of enzymes, which are selected for follow-up research. The cells continue to express the lacZ reporter gene product, beta-galactosidase derived from E. coli, so that individual tumor cells can be identified by histochemical staining of tissue sections. Lymphocytes and other responding cells on the same slide can also be identified by dual-labeled antibodies. The contrast between stained cells and background is useful for image analysis. This is one of the few microscopic brain tumor models that allow quantitative analysis. This tumor mimics the important characteristics of the growth and spread of human brain tumors. The expression of beta-galactosidase is very stable, but the cells should be cloned again after several months of culture.
