Description

β-secretase (β-position amyloid precursor protein (APP) cleaving enzyme 1 / BACE1) participates in the initial cleavage of amyloid precursor-APP, generating two soluble N-terminal and C-terminal fragments. Another secretase isoenzyme, Secretase, cleaves the C-terminal fragment to produce amyloid β. The accumulation of amyloid in the brain leads to the formation of amyloid plaques, which leads to the development of Alzheimer's disease (AD) in humans. Therefore, in Alzheimer's disease-related drug discovery research, the screening of new and specific human β-secretase inhibitors (BACE1) has become crucial. In the human β-secretase (BACE1) inhibitor screening kit for the determination of Genie, human BACE1 cleaves the BACE1 specific quenching substrate to produce a product with high fluorescence, which can be tested at Ex / Em = 435/500 Measured at nm. In the presence of a BACE1 inhibitor (control included in the kit), enzymatic cleavage is hindered and can be used to screen new inhibitor compounds.
