Description

β-secret enzyme (β-position amyloid precursor protein (APP) cleavage enzyme 1 / BACE1) participates in the initial cleavage of amyloid precursor-APP, generating two soluble N-terminal and C-terminal fragments. Another secretase isoenzyme γ-secretase cleaves the C-terminal fragment to produce amyloid β. The accumulation of amyloid β in the brain leads to the formation of amyloid plaques, which leads to the development of Alzheimer's disease (AD) in humans. Therefore, in Alzheimer's disease-related drug discovery research, the screening of new and specific human β-secretase inhibitors (BACE1) has become crucial. In our human β-secretase (BACE1) inhibitor screening kit, human BACE1 cleaves the specific quenching substrate of BACE1 to produce a highly fluorescent product that can be measured at Ex / Em = 435/500 nm. In the presence of a BACE1 inhibitor (the kit includes a control), enzymatic cleavage is hindered and can be used to screen new inhibitor compounds. The assay has high-throughput adaptability and can be completed in less than 1 hour.
