Target Overview

Presenilin-2 is a multi-pass transmembrane protein that is highly homologous to Presenilin-1. It serves as an essential catalytic subunit of the gamma-secretase complex, an intramembrane protease. This complex executes the final cleavage of the Amyloid Precursor Protein (APP) to generate different species of Amyloid-beta (Aβ) peptides. Mutations in the PSEN2 gene are a known cause of early-onset, inherited Alzheimer's disease (AD4), albeit less common than mutations in PSEN1. Its direct involvement in Aβ production makes it a key protein in the pathology of Alzheimer's.
