FUS (Fused in Sarcoma) is a DNA/RNA-binding protein that is predominantly located in the nucleus and plays a crucial role in various aspects of RNA metabolism, including transcription, pre-mRNA splicing, and RNA transport. FUS is strongly linked to neurodegenerative disease; mutations in the FUS gene are a known cause of familial Amyotrophic Lateral Sclerosis (ALS). In these diseases, a key pathological feature is the mislocalization of the FUS protein from the nucleus to the cytoplasm, where it forms insoluble aggregates. These FUS-positive inclusions are hallmarks of certain forms of ALS and Frontotemporal Lobar Degeneration (FTLD-FUS).
