Features

Each vial or HiTector M1 contains U2OS cells stably expressing unlabeled human muscarinic acetylcholine receptor M1. The Creative Biolabs M1 cell line is designed to test compounds or analyze their ability to modulate the muscarinic acetylcholine receptor M1. When the agonist binds to M1, it activates the G protein, which in turn triggers a cellular response mediated by the second messenger (calcium). This cell line has been validated by measuring the increase in calcium in the cytosol. The high reproducibility of this assay allows monitoring of the M1 activation process in high-throughput screening.

Assay Description

The M1 Muscarinic Acetylcholine Receptor Assay (CHRM1) from Creative Biolabs allows compounds to be tested or analyzed for their ability to modulate CHRM1. When the ligand binds to CHRM1, it activates the G protein; this in turn triggers calcium Mediated cell response. The combination of agonist and CHRM1 induces intracellular calcium mobilization and PKC activation through PLC-IP3 and DAG pathways, respectively.

The M1 muscarinic acetylcholine receptor test involves measuring the concentration of calcium in the cytoplasm. The high reproducibility of this assay allows monitoring the activation of CHRM1 receptors in living cells.
