Target Overview

Leucine-rich repeat kinase 2 (LRRK2) is a large, multi-domain enzyme that has both kinase and GTPase enzymatic functions. While its precise physiological roles are still being fully elucidated, LRRK2 is best known for its strong genetic association with Parkinson's disease. Mutations in the LRRK2 gene are the most frequent cause of both familial (inherited) and sporadic Parkinson's disease. These pathogenic mutations typically lead to a toxic gain-of-function by increasing the protein's kinase activity, which is thought to contribute to neuronal damage. This makes LRRK2 a primary therapeutic target for the development of kinase inhibitors.
