Description

This molecule is an advanced, genetically encodable RNA-based imaging tool. It consists of a precisely defined 49-nucleotide RNA sequence that folds into a stable and specific three-dimensional structure. Unlike therapeutic aptamers, its primary function is to act as a conditional fluorogenic module. By itself, the aptamer is non-fluorescent. However, upon binding its cognate small molecule fluorophore (such as DFHBI), it induces a conformational lock on the dye, causing it to become brightly fluorescent. This "light-up" mechanism provides a high signal-to-noise ratio for imaging.

Engineered for superior performance, this aptamer exhibits improved thermostability and a lower dependency on magnesium for proper folding compared to earlier iterations of fluorescent RNA tags. Crucially, it folds efficiently in vivo without the need for a stabilizing tRNA scaffold, making it a more robust and less intrusive tag for studying the life cycle of an RNA of interest.
