This is an engineered RNA motif that folds into a stable parallel G-quadruplex structure, creating a specific binding pocket for Thiazole Orange fluorophores. The aptamer exhibits exceptionally high binding affinity (low nanomolar range), leading to a massive fluorescence enhancement of over 1,000-fold. This makes it a superior tool for imaging low-abundance RNA and performing robust quantitative analysis in live cells.
